The SPI Test ™ (Sperm Pathogen Immunophenotyping Test) is a new, patented, award-winning diagnostic test that for the first time allows detection of intracellular pathogens such as viruses (e.g. CMV, HSV 1/2) and Chlamydia inside spermatozoa, by immunofluorescence and flow cytometry. The SPI™ TEST allows etiological investigation of male factor infertility, early pregnancy failure and recurrent miscarriages due to sperm-originating pathogens even in cases of chronic, subclinical infections where conventional methods such as cultures, immunofluorescence and PCR fail to detect them. It is the most sensitive test commercially available (more sensitive than PCR-based commercial tests), while at the same time it provides extreme specificity, allowing correlation between infection and infected cell type.
METHODOLOGY: Immunofluorecent analysis of semen sample using flow cytometry for the intracellular detection of microorganisms. Chlamydia (CT): Immunofluorecent analysis using a monoclonal antibody for the detection of Chlamydia (Chlamydia trachomatis). The antibody binds with a membrane protein of the Chlamydia and can identify all the 15 serotypes of the species Chlamydia trachomatis. A positive sample is considered by the relationship of fluorescence between the sample and the control; Cytomegalovirus (CMV): Immunofluorecent analysis using a monoclonal antibody for the detection of the human herpesvirus–5, (HHV–5 or CMV). The antibody reacts with immediate early (antigens expressed at 3-12 hrs post-infection generally involved in transcription such as 72kD major phosphoprotein), early (expressed at 12-24hrs post-infection, generally enzymes and one virion structural gene preceding viral DNA synthesis), and late antigen preparations (expressed at 36-48hrs post-infection, generally structural proteins). A positive sample is considered by the relationship of fluorescence between the sample and the control; Herpes Virus 1/2 (HSV 1/2): Immunofluorecent analysis using a monoclonal antibody for the detection of the human herpes simplex virus 1/2. The antibody recognises glycoprotein D (gD) of HSV 1 and 2. gD is a viral envelope component that is also expressed in the membranes of HSV infected cells. A positive sample is considered by the relationship of fluorescence between the sample and the control. The detection of the above infectious agents is assayed both on the membrane of the cells and secondly intracellular.

SAMPLE STORAGE & SHIPPING: After collection using our special solution containing double vial, samples should be stored at low temperature (4◦- 8◦) in a refrigerator (do not freeze) and shipped in our Fertility Kit box according to instructions at AMBIENT TEMPERATURE (no ice packs needed). Next day delivery (24-36 hours) is required (see instructions). Delivery of samples to Locus Medicus should be planned between Monday and Friday but not during the Weekend.
