Patient’s whole blood cell cultures are incubated in the presence of Ca+ for cells to deplete through exocytosis any cytokines already present in the cytoplasm. Whole blood is activated with PMA and ionomycin in the presence of a transport inhibitor and the produced cytokines after 4 hours incubation are detected with fluorochrome conjugated antibodies. Percentages of Th1 and Th2 cytokine-producing cells are then identified as the number of IFN-g-positive and IL-4-positive cells present, respectively, in the total population of CD4+ T-helper cells as determined by flow cytometry immunofluorescence analysis.

SAMPLE STORAGE & SHIPPING: Samples should be stored and shipped at ambient temperature and away from light. Next day delivery (24-36 hours) is required. Delivery of samples to our lab should be planned between Monday and Thursday but not Friday or during the Weekend. Sample collection and pick-up by courier should take place on the same day.
